112年度「實體講座:Targeting the DNA Damage Response in Myeloid Malignancy: From Genomes to Drugs」

講座摘要:
Dysregulation of the DNA damage response and p53 activation is a central mechanism by which normal and malignant hematopoietic cells survive genotoxic stresses such as chemotherapy and radiation. Our lab studies how PPM1D, a gene that is recurrently activated in clonal hematopoiesis and myeloid neoplasia, regulates normal hematopoietic stem cell function and how PPM1D contributes to the development of therapy resistance in myelodysplastic syndrome and acute myeloid leukemia. Using a combination of human genetics, mouse models, and engineered cell systems, we have found that PPM1D is a central regulator of hematopoietic cell fitness, particularly in the context of genotoxic stress, and is a target in myeloid malignancies. These biological insights have led to a drug discovery effort to target PPM1D and other regulators of the DNA damage response.
