Bioinformatics \u767c\u8868\u4e86\u4e00\u7bc7\u7814\u7a76\uff0c\u5229\u7528\u6f14\u7b97\u6cd5\u5c07\u4e0d\u540c\u76f8\u95dc\u7684\u958b\u653e\u8cc7\u6599\u5eab\u7d71\u6574\u5728\u4e00\u8d77\uff0c\u5efa\u7acb\u4e86\u7c92\u7dda\u9ad4\u6bd2\u6027\u7684\u5927\u6578\u64da\u8cc7\u6599\u5eabMitoTox\u3002MitoTox\u662f\u4e00\u500b\u63d0\u4f9b\u6bd2\u7269\u8cc7\u6599\u767c\u73fe\u8ddf\u56de\u5831\u7684\u958b\u653e\u5e73\u53f0\uff0c\u5229\u7528Python3\u690d\u5165\u591a\u500b\u4e0d\u540c\u7db2\u7ad9\u7684\u8cc7\u6599\u3002\u76ee\u524dMitoTox\u5305\u542b\u4e861400\u500b\u6df7\u5408\u9805\u76ee\u8207\u5176\u76f8\u5c0d\u61c9\u7684\u5171\u670934000\u500b\u4e0d\u540c\u7684\u540d\u8a5e\u3002\u9019\u6240\u6709\u7684\u9805\u76ee\u9084\u9023\u7d50\u4e86870\u591a\u7a2e\u7c92\u7dda\u9ad4\u6bd2\u6027\u76f8\u95dc\u7684\u86cb\u767d\u8cea\u3001\u86cb\u767d\u8cea\u526f\u7522\u7269\u8ddfRNA\u76ee\u6a19\u3002\u9019\u4e9b\u6bd2\u5316\u7269\u6839\u64da\u4ed6\u5011\u7684\u529f\u80fd\u548c\u6bd2\u7269\u6a5f\u5236\u88ab\u5206\u52308\u5927\u985e\u3001225\u529f\u80fd\u526f\u5206\u985e\u548c650\u500b\u76ee\u6a19\u5b50\u96c6\uff0c\u800c\u88ab\u8a18\u9304\u5728\u5176\u4e2d\u7684\u5168\u90e8\u9805\u76ee\u90fd\u6709\u9644\u4e0a\u4f86\u6e90\u3002<\/p>\n<p>\u50cf\u9019\u6a23\u9f90\u5927\u7684\u5de5\u7a0b\u4e5f\u6b78\u529f\u65bc\u73fe\u4eca\u8cc7\u6599\u79d1\u5b78\u7684\u6f14\u9032\uff0c\u800cMitTox\u7684\u5efa\u7acb\u4e0d\u50c5\u53ef\u4ee5\u77ad\u89e3\u7c92\u7dda\u9ad4\u7684\u6a5f\u5236(\u4f8b\u5982:\u7d30\u80de\u8cea\u4e2d\u7684\u96fb\u4f4d\u6539\u8b8a\u548cATP\u7684\u88fd\u9020\u904e\u7a0b)\u66f4\u53ef\u4ee5\u4fc3\u9032\u7528\u85e5\u5b89\u5168\u4ee5\u53ca\u6bd2\u6027\u9810\u9632\u3002\u5982\u6b64\u9ad8\u541e\u5410\u91cf\u7684\u8cc7\u6599\u7cfb\u7d71\uff0c\u63d0\u4f9b\u4e86\u7d71\u8a08\u5206\u6790\u8ddf\u6a5f\u5668\u5b78\u7fd2\u4f86\u7814\u7a76\u7c92\u7dda\u9ad4\u7684\u904b\u4f5c\u6a5f\u5236\uff0c\u4e5f\u53ef\u4ee5\u4fc3\u9032\u672a\u4f86\u767c\u73fe\u6216\u9810\u6e2c\u66f4\u591a\u7684\u7c92\u7dda\u9ad4\u6bd2\u7269\u53ca\u6cbb\u7642\u7684\u65b9\u6cd5\u3002\u5728\u6587\u672b\u9b4f\u8001\u5e2b\u4e5f\u63d0\u5230\uff0cMitoTox\u7684\u521d\u8877\u4fbf\u662f\u70ba\u4e86\u5b78\u7fd2\u85e5\u7269\u662f\u600e\u9ebc\u5f15\u767c\u7c92\u7dda\u9ad4\u7522\u751f\u6bd2\u6027\u4ee5\u53ca\u5176\u4ed6\u526f\u4f5c\u7528\u7684\u5f71\u97ff\u3002MitoTox\u5efa\u8d77\u4e86\u7c92\u7dda\u9ad4\u6bd2\u6027\u8ddf\u526f\u4f5c\u7528\u4e4b\u9593\u7684\u6a4b\u6a11\uff0c\u4e5f\u70ba\u4e86\u672a\u4f86\u7684\u91ab\u5b78\u9810\u6e2c\u63d0\u4f9b\u4e86\u4e00\u689d\u6377\u5f91\u3002<\/p>\n<p><img structure<\/p>\n<p>&nbsp;<\/p>\n<p>&nbsp;<\/p>\n<p>The mitochondria are the energy factories of the human cells responsible for cellular metabolism and the production of adenosine triphosphate (ATP). Mitochondria play a crucial role in regulating cellular functions, and they can be damaged and produce side effects when exposed to external drugs or molecules. Therefore, it is essential to construct a comprehensive database of big data that correlates different drugs or molecules with their corresponding side effects on cells. This not only helps doctors better diagnose patients&#8217; symptoms but also allows us to further understand the working principles of mitochondria.<\/p>\n<p>&nbsp;<\/p>\n<p>Mitochondrial dysfunction is often associated with cardiovascular diseases, neurodegenerative disorders (such as Alzheimer&#8217;s), and cancer. Therefore, studying the impact of mitochondria on human health is of paramount importance. Dr. Wei from our institute is dedicated to mitochondrial research. In 2021, Dr. Wei published a study in BMC Bioinformatics, in which an algorithm was used to integrate various relevant open databases, creating a big data database called MitoTox, which focuses on mitochondrial toxicity. MitoTox is an open platform for discovering and reporting toxicological data, using Python3 to extract data from multiple websites. Currently, MitoTox includes 1,400 mixed items with 34,000 different terms associated with them. All these items are linked to over 870 mitochondrial toxicity-related proteins, protein by-products, and RNA targets. These toxins are categorized into eight major classes, 225 functional subcategories, and 650 target subsets, with their sources provided for all recorded items.<\/p>\n<p>&nbsp;<\/p>\n<p>The establishment of a project of this magnitude is made possible by the evolution of data science today. The creation of MitoTox not only enhances our understanding of mitochondrial mechanisms (such as changes in cytoplasmic potential and ATP production processes) but also promotes drug safety and toxicity prevention. This high-throughput data system provides statistical analysis and machine learning tools to study the functioning mechanisms of mitochondria and facilitates the discovery and prediction of more mitochondrial toxins and treatment methods in the future. In conclusion, Dr. Wei also mentioned that the initial intention of MitoTox was to learn how drugs induce mitochondrial toxicity and other side effects.
