therapy\uff0cRT\uff09\u4f86\u6cbb\u7642\u3002\u4f46\u5148\u524d\u7684\u7814\u7a76\u767c\u73fe\u900f\u904e\u653e\u5c04\u6cbb\u7642\u6cbb\u7642\u808c\u8089\u4fb5\u72af\u578b\u8180\u80f1\u764c\u53ef\u80fd\u6703\u56e0\u70ba\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\uff08Tumor-associated macrophage\uff0cTAM\uff09\u7684\u53c3\u8207\u4ee5\u4fc3\u9032\u764c\u75c7\u7d30\u80de\u7684\u8f49\u79fb\uff0c\u65bc\u662f\u6210\u6559\u6388\u8207\u5176\u5718\u968a\u5247\u5e0c\u671b\u53ef\u4ee5\u6df1\u5165\u7814\u7a76\u653e\u5c04\u6cbb\u7642\u8207\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u9593\u7684\u4f5c\u7528\u95dc\u4fc2\uff0c\u4ee5\u89e3\u958b\u5169\u8005\u4e4b\u9593\u975e\u9810\u671f\u4fc3\u9032\u8f49\u79fb\u7684\u4f5c\u7528\u6a5f\u5236\uff0c\u4f86\u63d0\u5347\u653e\u5c04\u6cbb\u7642\u7684\u6548\u7528\u3002<\/p>\n<p>&nbsp;<\/p>\n<p>\u6210\u6559\u6388\u8207\u5176\u7814\u7a76\u5718\u968a\u70ba\u63a2\u8a0e\u63a5\u53d7\u653e\u5c04\u6cbb\u7642\u7684\u8180\u80f1\u764c\u7d30\u80de\u8207\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u7684\u4ea4\u4e92\u4f5c\u7528\uff0c\u4ed6\u5011\u9996\u5148\u5efa\u7acb\u4e86\u7570\u4f4d\u79fb\u690d\u5c0f\u9f20\u6a21\u578b\u4f86\u8a55\u4f30\u653e\u5c04\u6cbb\u7642\u7684\u6548\u679c\uff0c\u4ed6\u5011\u767c\u73fe\u5c40\u90e8\u7684\u653e\u5c04\u6cbb\u7642\u6709\u52a9\u65bc\u6291\u5236\u5c0f\u9f20\u7684\u816b\u7624\u751f\u9577\uff0c\u4f46\u537b\u4e5f\u51fa\u73fe\u4e86\u80ba\u90e8\u8f49\u79fb\u7684\u73fe\u8c61\u3002\u800c\u70ba\u4e86\u63a2\u7a76\u8180\u80f1\u764c\u7d30\u80de\u8207\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u7684\u76f8\u4e92\u4f5c\u7528\u95dc\u4fc2\uff0c\u4ed6\u5011\u4f7f\u7528Transwell\u5171\u57f9\u990a\u7cfb\u7d71\u4f86\u6a21\u64ec\u9ad4\u5167\u74b0\u5883\uff0c\u900f\u904e\u8a72\u5be6\u9a57\u7cfb\u7d71\uff0c\u4ed6\u5011\u767c\u73fe\u5728\u7d93\u904e\u653e\u5c04\u7dda\u7167\u5c04\u7684\u8180\u80f1\u764c\u7d30\u80de\u6703\u589e\u52a0CCL2\u7684\u5206\u6ccc\uff0c\u9023\u7d50\u5230\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u4e0a\u7684CCR2\u53d7\u9ad4\uff0c\u4fc3\u4f7f\u5176\u81ea\u5177\u6709\u6291\u5236\u764c\u75c7\u751f\u9577\u7684M1\u985e\u578b\u5de8\u566c\u7d30\u80de\uff08M1-type TAM\uff09\uff0c\u8f49\u8b8a\u6210\u5177\u6709\u6291\u5236\u514d\u75ab\u7cfb\u7d71\u529f\u80fd\u3001\u4fc3\u9032\u764c\u75c7\u7d30\u80de\u751f\u9577\u7684M2\u985e\u578b\u5de8\u566c\u7d30\u80de\uff08M2-type TAM\uff09\uff0c\u4f7f\u8180\u80f1\u764c\u7d30\u80de\u53ef\u4ee5\u8eb2\u904e\u514d\u75ab\u7cfb\u7d71\u7684\u4fb5\u8972\uff0c\u4e26\u4e14\u8f49\u79fb\u81f3\u5176\u4ed6\u5668\u5b98\u3002\u6700\u5f8c\u6210\u6559\u6388\u7684\u5718\u968a\u4e5f\u900f\u904e\u52a0\u5165CCR2\u62ee\u6297\u5291\uff0c\u963b\u65b7CCL2-CCR2\u7684\u6d3b\u5316\u3002\u4ed6\u5011\u767c\u73fe\u6b64\u65b9\u6cd5\u53ef\u4ee5\u6709\u6548\u963b\u6b62\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u81eaM1\u985e\u578b\u8f49\u8b8a\u6210M2\u985e\u578b\uff0c\u4e26\u4e14\u4e5f\u6291\u5236\u80ba\u90e8\u7684\u764c\u75c7\u8f49\u79fb\u3002\u7d9c\u5408\u4ee5\u4e0a\u5be6\u9a57\u6210\u679c\uff0c\u6210\u6559\u6388\u767c\u73fe\u589e\u52a0CCL2\u7684\u8868\u73fe\u662f\u8180\u80f1\u764c\u7d30\u80de\u5c0d\u653e\u5c04\u6cbb\u7642\u7684\u53cd\u61c9\u4e4b\u4e00\uff0c\u975e\u9810\u671f\u589e\u52a0\u4e86M2\u985e\u578b\u7684\u816b\u7624\u76f8\u95dc\u5de8\u566c\u7d30\u80de\u4f86\u5275\u9020\u514d\u75ab\u6291\u5236\u7684\u74b0\u5883\uff0c\u4f7f\u764c\u75c7\u7d30\u80de\u53ef\u4ee5\u66f4\u4e0d\u53d7\u514d\u75ab\u7cfb\u7d71\u63a7\u5236\uff0c\u4e5f\u4fc3\u9032\u5176\u8f49\u79fb\u6f5b\u529b\uff0c\u9019\u500b\u7406\u8ad6\u63d0\u4f9b\u81e8\u5e8a\u6cbb\u7642\u8180\u80f1\u764c\u6642\uff0c\u4f7f\u7528\u653e\u5c04\u6cbb\u7642\u5fc5\u9808\u6ce8\u610f\u907f\u514d\u7684\u975e\u9810\u671f\u73fe\u8c61 [1]\u3002<\/p>\n<p>&nbsp;<\/p>\n<p>\u9664\u4e0a\u8ff0\u7684\u7814\u7a76\u6210\u679c\u5916\uff0c\u6210\u6559\u6388\u4e5f\u81f4\u529b\u65bc\u809d\u764c\u7684\u76f8\u95dc\u7814\u7a76\u3002\u8fd1\u671f\u6210\u6559\u6388\u7684\u4e00\u7bc7\u7814\u7a76\u6210\u679c\u4e2d\u767c\u8868\u4e86\u653e\u5c04\u6cbb\u7642\u8207\u7d44\u86cb\u767d\u53bb\u4e59\u91af\u5316\u9176\u6291\u5236\u5291\uff08Histone deacetylase inhibitor\uff0cHDACi\uff09\u7684\u5408\u4f75\u4f7f\u7528\u6709\u52a9\u65bc\u63d0\u5347\u6297\u764c\u4f5c\u7528\u7684\u6f5b\u5728\u539f\u56e0\u3002\u6210\u6559\u6388\u900f\u904e\u5c0f\u9f20\u6a21\u578b\u8207\u9ad4\u5916\u7d30\u80de\u57f9\u990a\u7684\u5be6\u9a57\u767c\u73fe\u653e\u5c04\u7dda\u7d50\u5408HDACi\u7684\u6cbb\u7642\u7b56\u7565\u53ef\u4ee5\u589e\u52a0\u809d\u764c\u7d30\u80de\u4e2dNKG2D\u914d\u9ad4\u7684\u8868\u9054\uff0c\u4f86\u4f7f\u5f97\u4ed6\u5011\u6703\u66f4\u5bb9\u6613\u53d7\u5230\u81ea\u7136\u6bba\u624b\uff08Natural [3]\u3002<\/p>\n<p>&nbsp;<\/p>\n<p>Research Topic: Exploring combination strategies of radiation therapy to enhance treatment efficacy.<\/p>\n<p>&nbsp;<\/p>\n<p>Bladder cancer is a common malignant disease in the urinary system, and it is more prevalent in middle-aged and elderly males. When cancer cells extend from the bladder mucosal layer to the muscle layer, it is clinically classified as muscle-invasive bladder cancer (MIBC). In addition to surgery and chemotherapy, radiation therapy (RT) is also used to treat this type of bladder cancer. However, previous studies have found that RT for MIBC may promote cancer cell metastasis through the involvement of tumor-associated macrophages (TAMs). Therefore, Professor Cheng and his team aim to investigate the interactions between radiation therapy and tumor-associated macrophages to uncover the underlying mechanisms that contribute to tumor metastasis and enhance the efficacy of radiation therapy.<\/p>\n<p>&nbsp;<\/p>\n<p>Professor Cheng and his research team aimed to investigate the interaction between irradiated bladder cancer cells and TAMs. They first established an ectopic allograft mouse model to evaluate the therapeutic effects of radiation therapy. They found that local RT delayed tumor growth in mice but also led to pulmonary metastases. To study the interaction between bladder cancer cells and TAMs, they used Transwell coculture system to simulate the <em>in vivo<\/em> environment. Through this experimental system, they discovered that irradiated bladder cancer cells increased the secretion of C-C motif chemokine ligand 2 (CCL2), which bound to C-C motif chemokine receptor 2 (CCR2) on TAMs. This interaction caused the polarization of TAMs from the M1 anti-tumor type to the M2 pro-tumor type, promoting bladder cancer cell invasion and evasion of the immune system. In the final step, Professor Cheng&#8217;s team added a CCR2 antagonist to block the activation of CCL2-CCR2. They found that this approach effectively prevented the phenotypic transformation of TAMs from M1 to M2 and inhibited cancer metastasis to the lungs. Based on these experimental findings, Professor Cheng discovered that increased expression of CCL2 is one of the responses of bladder cancer cells to RT, leading to the recruitment of M2-type TAMs and the creation of an immunosuppressive environment that allows cancer cells to evade immune control and enhances their metastatic potential [1].<\/p>\n<p>&nbsp;<\/p>\n<p>In addition to the aforementioned research findings, Professor Cheng is also dedicated to conducting the related studies on liver cancer. In a recent research publication, Professor Cheng demonstrated that the combination of RT and histone deacetylase inhibitor (HDACi) could potentially enhance the anticancer effects. Through experiments conducted on mouse models and <em>in vitro<\/em> cell cultures, Professor Cheng found that the treatment strategy combining RT with HDACi increased the expression of NKG2D ligands in liver cancer cells, making them more susceptible to targeting and killing by natural killer cells [2]. Another publication explored the impact of targeted monotherapy, combined targeted drug therapy, and immunotherapy on patients with inoperable liver cancer. They found that, compared to single-targeted therapy, the combination of targeted drug therapy and immunotherapy significantly improved patients&#8217; survival rates [3].<\/p>\n<p>&nbsp;<\/p>\n[1]\u00a0\u00a0 Chiang, Y., Tsai, Y.
